Lab Report: Cell Cycle Analysis in Onion Root Tip Cells

Objective:
To determine the percentage of time spent in each phase of the cell cycle (Interphase
and Mitosis) in onion root tip cells, and to calculate the total time spent in each phase
based on cell cycle data.

Background:The cell cycle consists of a series of events that lead to cell division. It can
be broken into two main phases: Interphase and Mitosis. Mitosis itself is divided into
several stages: Prophase, Metaphase, Anaphase, and Telophase. The duration of each
phase varies among different types of tissues, but certain general trends are known:

- Prophase is typically the longest phase (1-2 hours).
- Anaphase is the shortest phase (2-10 minutes).
- Metaphase (5-15 minutes) and Telophase (10-30 minutes) are also relatively short.
- Interphase can range from 12 to 30 hours, and it is the phase during which the cell
prepares for division.

In this experiment, the time spent in each phase was calculated using the percentage of
cells observed in each phase during the onion root tip cell cycle, assuming an average
total cell cycle time of 960 minutes (16 hours).

Hypothesis:
Based on known cell cycle data for onion root tip cells, we hypothesize that:
- Most of the time in the cell cycle will be spent in Interphase.
- Mitosis, specifically Prophase, will be the shortest overall period.

Materials:
- Onion root tips
- Microscope slides
- Methylene blue stain (for cell observation)
- Microscope
- Timer
- Graduated pipette

Procedure:
1. Preparation of Onion Root Tips:
- Onion root tips were collected and stained with methylene blue to highlight the
chromosomes.

, - The root tips were then observed under the microscope to identify the stages of the
cell cycle (Interphase, Prophase, Metaphase, Anaphase, Telophase).

2. Cell Observation:
- Three different fields of view were selected on the microscope slide.
- The number of cells in each phase was recorded for each field (Field 1, Field 2, Field
3).

3. Data Collection:
- The number of cells in each phase was recorded for each field, and the total number
of cells across all fields was calculated.
