a. Glycerolipiden
Oude membraanlipiden moeten constant vervangen worden door nieuwe en groeiende cellen. Of
wanneer de totale membraanoppervlakte moet uitbreiden zijn ook nieuwe lipiden nodig.
- Glycerolipiden: glycerol + 2 vetzuren veresteren
- Sfingolipiden: sfingosine + 1 vetzuur veresteren
- Cholesterol: geen verestering
De novo synthese van glycerolipiden
Hij begint gemeenschappelijk met de synthese van triglyceriden
→ synthese fosfatidaat.
1. Productie van CDP-diacylglycerol, door CTP
2. Dit reageert met OH-groep van serine, ethanolamine,
choline of inositol. CMP wordt gevormd.
Fosfatidylserine → fosfatidylethanolamine door decarboxylering
Fosfatidylethanolamine → fosfatidylcholine door 3x methylering
Salvage synthese van glycerolipiden
Hergebruiken van vrij choline, dat eerst door cholinekinase wordt gefosforyleerd tot fosforylcholine
en dan wordt omgezet tot CDP-choline (≈UDP-glucose). Dit condenseert dan met diacylglycerol zodat
fosfatidylcholine ontstaat. Voor ethanolamine is er een gelijkaardige weg.
Toepassing: de moleculaire verklaring voor surfactans
Dipalmitoyllecithine is een tensoactieve stof die de oppervlaktespanning tussen water en lucht verlaagt →
longfysiologie, alveolen hebben een groot oppervlak met lucht. Er zijn 2 soorten alveolen.
1) Type I alveolaire cellen (staan in voor gasuitwisseling).
Tussen membraan van type I en lucht zit er een dunne laag moleculen → brug tussen lucht en
waterige fase van cellen = surfactans. Bestaat vooral uit dipalmitoyllecithine.
Bestaat ook uit proteïnen die zorgen voor immuunsysteem want ze reageren met microben →
gaan fagocytose bevorderen.
2) Type II alveolaire cellen (produceren surfactans). Zonder surfactans treedt er een collaps op van
alveolen → contactoppervlak neemt af → ademhalingsproblemen. Dit komt voor bij baby’s →
acuut respiratoir distress-syndroom (ARDS) hebben te weinig surfactans.
, b. Sfingolipiden
De novo synthese sfingolipiden
1. Reactie tussen palmitoyl-CoA en serine.
2. Dubbele redoxreactie die een tweede alcoholgroep en een dubbele binding maakt
3. Aminoalcohol wordt veresterd met vetzuur → ceramide
Hieruit vertrekken 2 wegen:
CDP-geactiveerd choline UDP-geactiveerd glucose/galactose
→ sfingomyeline → cerebrosiden
→ + UDP -geactiveerde suikers
→ gangliosiden (oligosachariden)
Deze gangliosiden zijn geladen en die komen in contact met waterfase van extracellulaire ruimte. Er
zijn veel verschillende gangliosiden met veel variatie (bv: ABO-bloedgroepen). Vooral in neuronen
want ze spelen rol in signaaltransductie.
Toepassing: stapelingsziekten van sfingolipiden
Ziekte van Tay-Sachs en ziekte van Sandhof: Bèta-hexosaminidase ontbreekt. GM2gangliosiden stapelen
zich op in lysosomen van neuronen en deze zwellen op. → psychomotorische achterstand, blindheid
(degeneratie netvlies), en rood oog.
Ziekte van Gaucher: ontbreken van bèta-glucocerebrosidase zodat glucocerebrosiden zich opstapelen in
fagocyten van reticulo-endotheliale systeem → overdreven werkende milt, vermindering van
bloedplaatjes, botpijn, neurologische stoornissen.
Ziekte van Niemann-Pick: sfingomyelinase ontbreekt en sfingomyeline stapelt op in lysosomen →
achterstanden in psychomotorische ontwikkeling, lever en bloed.
