Probleem: Cellen zijn gemaakt uit heel veel verschillende moleculen. We willen 1 bepaald type
lokaliseren, maar hoe doen we dit?
Oplossing: Immunofluorescentie om specifieke cel componenten te identificeren. Fluorescent
molecule wordt aan een antilichaam vastgemaakt dat bindt aan de bepaalde molecule die we willen
onderzoeken. Wanneer we dan een fluorescente microscoop gebruiken kunnen we deze moleculen
lokaliseren.
Antilichaam: proteïne die bestaat uit:
- constante regio (hetzelfde voor alle antilichamen)
- Variabele regio (identiek aan elkaar, maar uniek voor elk antilichaam)
- V-regios waarin een specifiek antigen past.
Kunnen gegenereerd worden in een laboratorium door een
onbekende proteïne of macromolecule in te spuiten in muizen of
konijnen, die hierna antilichamen gaan produceren die bindt aan
welke proteïne dan ook.
Primaire immunofluorescentie:
Antilichamen zijn gelabeld met een fluorescente kleurstof
(fluorophore) die covalent gebonden is aan een C-regio binnenin het
antilichaam.
Secundaire immunofluorescentie:
Cel wordt behandeld met niet-gelabelde antilichamen
(primaire antilichamen), deze gaan binden aan het target molecule.
Wel-gelabelde antilichamen (secundaire antilichamen) komen daarna
in de cel en binden aan de primaire antilichamen.
Er kunnen meerdere primairen aan een target binden en meerdere
secundaire aan een primaire, dus er zal veel duidelijker beeld zijn.
