Lab 6: Genetic Identification of Bacteria

The purpose of the lab is to familiarize you with the science and techniques (specifically, the BLAST
technique) used to identify different types of bacteria based on their DNA sequence. Not long ago, DNA
sequencing was a time-consuming, tedious process. With readily available commercial equipment and
kits, it is now routine. The techniques used in this lab are applicable in a wide variety of settings,
including scientific research and forensic labs. . Go to the lab site (You will need to make sure Adobe
Shockwave plugin is installed on your computer so the lab will run correctly.)

Start the lab by clicking on “Click to enter the lab”. Start with the instructions on “Sample
Preparation”. Follow the steps and prompts answering quiz questions when directed. After you have
completed all the sections and quizzes, answer the questions below. After you identify Sample A, be
sure to follow the instructions on how to identify the remaining Samples B-F using the BLAST data
output and the NCBI website. Concentrate on increasing your comprehension of using genetic
techniques to identify organisms.

After each sample (A-F), you will have the option to open a window showing the data output from your
sequencer. For each sample (A-F), take a screenshot of this window showing your gene sequence for
each sample (the sequence that you run through the NCBI website), paste it into a document and save
it in PDF format, and upload it in the appropriate place as indicated below. All 6 gene sequences for
each sample should be submitted as a single PDF document and CLEARLY LABELLED.
