What the Body Does to the Drug

Pharmacokinetics deals with the movement of drugs through the body, broken
down into four core phases (ADME).

Absorption

Definition: Process of a drug moving from its site of administration into
the bloodstream.
Bioavailability (F): The fraction of an administered drug dose that
reaches systemic circulation unchanged.
IV Route: 100% bioavailability (F = 1.0).
Oral Route: <100% due to incomplete absorption and first-pass
metabolism.
First-Pass Effect: Oral drugs pass through the GI tract into the portal
vein and are metabolized by the liver before entering systemic
circulation.

doses compared to intravenous doses.



Distribution

Definition: Reversible transfer of a drug from one location (blood) to
another within the body.
Volume of Distribution(Vd): Fluid volume required to contain the entire
