Purification Of Gfp Fusion Proteins With Imac In E. Coli Cells
PURIFICATION OF GFP FUSION
PROTEINS WITH IMAC IN E.
Coli CELLS

ABSTRACT
This report was performed to demonstrate
techniques of purifying Green Fluorescent
Protein (GFP). The main technique used for
demonstration is by Immobilized Metal
Affinity Chromatogrpahy (IMAC). The
open reading frame for enhanced green
fluorescent protein was subcloned into the
pQE30 expression. The subsequent
production of protein tagged N-terminally
with hexahistidine, facilitated the
purification by IMAC

Identification Of Recombinant Gfp Protein Using Cloning, Expression And Purification
IDENTIFICATION OF RECOMBINANT GFP PROTEIN USING CLONING, EXPRESSION
AND PURIFICATION

Abstract. 144
Green fluorescent protein is an useful tool to
observe the presence of protein-protein
interaction as well as gene expression(Chalfie
et al., 1994). Which those experiment include
amplification and recombination of GFP open
reading from using RT-PCR which can clone
GFP and insert into pQE30 vector with
IPTG(Riedy et al., 1995). which is used to
promote expression of N terminal
hexahis...

Summary The Importance of shapes fitting together in cells and organisms - Paper 3 essay
ENZYMES:
-lock + key vs induces fit
-Reasons for enzymes specificity (TS) - -Formations of E-S complexes = main e.g. of
imp of complementary fitting
-Initial hypothesis = lock + key (only one specific mol binds-enzyme)
-But more recently accepted hypothesis = induced fit:
-because found that sim mols could still = complementary + bind to AS region of an
enzyme (∴→e-s)
-As complex = made, it allows a conformational change (by stretching + compressing
bonds of sub) ∴ reaction can oc...

IB Bio Ch. 3 & 7 (Enzyme Activity)
3.6.1 Define enzyme and active site - Enzymes: globular protein functioning as biological
catalyst, speeding up reaction rates by lowering activation energy.
Active site: site on the surface of enzyme to which substrate binds
3.6.2 Explain enzyme-substrate specificity and the relationship - Globular enzyme has a
specific three-dimensional shape active site that is complementary to the shape of
specific substrate
