H1: UV/VIS spectrofotometrie
1.1. Algemeen: spectrochemie




Elektromagnetische straling (EMS)
- Golflengte =  frequentie, , f amplitude
- Energie = h *  = h * c/ 
- Hoge golflengte = kleine E kleine golflengte = grote E → omgekeerd evenredig
Eigenschappen EMS
- Polarisatie: met rooster
- Breking: glaswater met pen erin
- Dispersie: wit licht → prisma → regenboog
- Interferentie: golflengten in fase = versterken niet in fase = doven
Interactie EMS en materie
- Bij UV: valentie-elektronen + straling → springen naar hogere schil
- Bij IR: niet genoeg energie om te laten springen, atomen trillen id moleculen
- Bij radiogolven: kern draait rond eigen as → bij genoeg energie → veranderen richting vd

1.2. Inleiding
Spectrometrie van UV/VIS → meest toegepaste techniek
Golflengtebereik:
- UV: 200 - 400 nm
- VIS = 400 – 700/800 nm
De kleur van de oplossing is de kleur die NIET wordt geabsorbeerd
- Geabsorbeerde kleur en doorgelaten kleur → complementair
- Vb. malachietgroen
o Absorbeert beste rood en oranje: 620 nm
o 450-500 nm → niet geabsorbeerd




1.3. Kwantitatieve aspecten metingen
Principe:
- M + h* → M*
o M = deeltje molecule in grond
o H* = EMS, energie
o M* = aangeslagen

- M* → M + energie (warmte)

1.3.1.1. Kwantitatieve aspecten
- Absorbantie = A → licht geabsorbeerd
- Transmissie, %, T → licht doorgelaten
- Concentratie: hoe donkerder het kleur vd oplossing, hoe hoger

vb. als 10 I van 100 I0 doorgelaten wordt → dan is de transmissie 10%

Transmissie en concentratie zijn omgekeerd evenredig
