Hoe kunnen we van complexe aandoeningen, dus van niet Mendeliaanse aandoeningen de
suspectibiliteitsgenen en de varianten achterhalen.

18.1 Investigation of complex disease: epidemiological approaches

rechts en de drempelwaarde blijft hetzelfde, het fenotype loopt in de familie.

Hoe kunnen we aantonen dat er onderliggende genen zijn voor die genetische aandoeningen.
Hoe kunnen we de genetische bijdrage aan het fenotype bepalen.
Adhv familie, tweeling en adoptie studies.
Hiervoor moeten we eerst criteria definieren die personen merken als aangetast of niet. Hier
gaan we niet verder op in.

, Families gebruiken om te achterhalen of er een genetische bijdrage is aan de ontwikkeling van
de niet mendeliaanse aandoening.

Als een aandoening mee genetisch bepaald wordt dan verwacht je dat bloedverwanten van een
aangetastte persoon meer risico hebben om het fenotype te ontwikkelen omdat ze genen met
elkaar gemeen hebben, hun suspectibiliteit schuift op naar rechts en de drempelwaarde blijft
hetzelfde, het fenotype loopt in de familie, er is familiale clustering.
De risk ratio is een maat voor familiale clustering.
Wanneer de risk ratio gelijk is aan 1 is er evenveel riscio bij de bloedverwanten als bij de
gewone populatie.
