DEEL I: INLEIDING TOT DE CELBIOLOGIE
1. Ontstaan van eukaryote cellen
1e organismen: foto-autotrofie: productie eigen voedsel door fotosynthese (kan wel/ geen O 2 productie doen)
fotosynthese: nCO 2 + nH2O + Ezonlicht → (CH2O)n + nO2
▪ Prokaryoten: archaea & bacteria Eukaryoten
* Geen kern: DNA (cirkelvormig chromosoom) los in de cel * Kern met chromosomen
* Celmembraan en celwand * Celmembraan (bij dieren!)
* Eéncellig * Eén/meercellig
* Niet gecompartimenteerd * Organellen
* Deling door doorsnoering * Deling mitose/meiose
* Heeft voorloper * Celwand (bij planten/ fungi)

Eukaryote cellen:

- Groter V door interne cytomembranen waaronder de kernenvelop (kernmembraan) met DNA.

- Cytoskelet met bijhorende motorische systemen→ celcompartimentering en spontane beweging.
- Organisatie genoom & ruimtelijke scheiding van transcriptie en transitie→ diversificatie.
- Mitochondria, peroxisomen & plastiden (planten)→ interiorisatie respiratie & energieproductie.



2. Chemische bouwstenen van cellen
Eiwitten, nucleïnezuren, polysachariden: polymeren, samengesteld uit monomeren
▪ Eiwitten= proteïnen, bestaande uit vele AZ (enkel L-vormen)
- Korte ketens (weinig AZ)= peptiden
- Langere keten (veel AZ)= polypeptiden
- Residu= individueel AZ in keten

 25 AZ gebruikt voor eiwitsynthese:
o Centraal koolstofatoom
o 4 verschillende chemische groepen: amino (op einde= C-terminus), carboxyl(op einde= N-
terminus), waterstof, variabele R groep(belangrijk voor opvouwing)

 Primaire structuur: bepaalt specifieke opeenvolging AZ/ polypeptideketens
 Secundaire structuur: ontstaan door opvouwing gelokaliseerde gedeelten <AZ (soms met hulp
<moleculaire chaperones)
 Tertiaire structuur: conformatie heel eiwit (gebeurt spontaan)

Conformatie: spontaan, soms bijgestaan door moleculaire chaperones
Denaturatie= verbreken natuurlijke conformatie door bv. temperatuurverhoging ( renaturatie)

AZ: polair, apolair, zuur of base
▪ Nucleïnezuren (zie bouw DNA)

▪ Polysachariden
bouwstenen:
- Monosachariden= koolhydraten bv. glucose (eenvoudigste suikers)
- Disachariden bv. lactose, maltose, sucrose (eenvoudigste polysachariden)
- Polysachariden bv. glycogeen, zetmeel, cellulose

* Glycoproteïnen= eiwitten met covalent gebonden koolhydraatketens
* Proteoglycanen= eiwit-koolhydraat complex met gespecialiseerde polysacharideketens
bv. glycosaminoglycanen= GAGs

▪ Vetten= lipiden
1) Vetzuren: CH-keten+ -COOH: verzadigd(geen dubbele b)/ onverzadigd(1 dubbele b)
2) Triglyceriden: triacylglycerols: glycerol + 3 vetzuurketens
3) Fosfolipiden: fosfoglyceriden & sfingolipiden
4) Glycolipiden: vgl. fosfolipiden met koolhydraat- i.p.v. fosfaatgroep
5) Steroïden: cholesterol (uitgangspt. steroïde hormonen)
6) Terpenen: vb carotenoïden

- Hydrofobe staart+ hydrofiele kop=> bepalen membraansamenstelling
- Onverzadigde vetten: belangrijk voor mens (hoe meer, hoe lopender de celmembraan)



3. Energiehuishouding in de cel
Cel heeft nodig: energie +chemische bouwstenen (foto/chemotrofen)

- Chemotrofen: voedsel opgenomen door mechanismen: sommige bacteriën, algen, planten, …
- Fototroof: voedsel geproduceerd door mechanismen: fungi, meeste bacteriën, dieren, …

Metabolisme= geheel <alle chemische reacties in cel (bestaat uit metabole pathways):

- Anabolische pathways: verbruiken E+ opbouw celcomponenten
- Katabolische pathways: leveren E+ afbraak celcomponenten (deze E w dan in anabolische pathways gebruikt)

Molecule voor stockage E= ATP (adenosine trifosfaat) ---→ ADP + fosfaatgroep ATP + H2O
