If you are using a compound microscope with an ocular lens with a magnification of 10x and an objective lens with a magnification of 40x, what is the total magnification? 400x 4. In this lab, we discussed preparation of a wet mount slide. Research and describe another slide preparation and an example of when it would be used. Dry mount slide: Put a few drops of liquid in the concavity and add water to thin it out if necessary. A dry mount slide can be prepared in a very similar way to wet mounts, except without the water. Place the object in the middle of the slide, and holding the cover slip at an angle, carefully set it on top. Experiment 1 Post-Lab Questions 1. What is the first step normally taken when you look through the ocular lenses? The first step when looking through the ocular lenses is adjusting the eye pieces to line up the light is the first step. 2. What does it mean that the image is inverted when you look through the ocular lenses? The image is mirrored to the person looking at it. An example of which is if you look at a picture and flip the right side to be on the left side and vice versa for the left side. 3. What new details are you able to see on the slide when the magnification is increased to 10x that you could not see at 4x? What about at 40x? On 10x went from the Large clump of a mass to a much more detailed view including the nucleus. The 40x was even more detailed and I saw smaller dots and th lines through the membrane. 4. What were your calibration measurements for the following objectives? 4x: 4.4 mm 10x: 2.1 mm 40x: .5 mm 5. What were your objective and measurement estimates for the following specimens? Sand dollar juvenile: 40x .3 mm Sand dollar larva: 10x 1.2 mm Sand dollar embryo: 40x .1 mm Human hair: 40x .1 mm Blade of grass: 4x 4.2 mm 6. Why is it important to be able to properly calibrate and measure objects viewed through a microscope? If you don’t pay attention to what calibration, then it will be hard to tell the size of the object and the length of the object at the proper magnification. 7. Describe the qualitative difference you notice with the different types of microscope views in the “Microscope Compare” and “Specimen Compare” exercises. The Microscope compare gives more detail, higher zoom and color. The Specimen compare gives much more variance in zoom.
