Carbohydrates are the most common organic substances found in nature. This
macromolecule is contained in both plants and animals, and they provide man with
needed energy for survival.

The isolation of glycogen from chicken or pork liver is attained by using the
mechanism of precipitation


ISOLATION OF LIVER GLYCOGEN

Materials:

Fresh chicken or pork liver
95% Ethyl alcohol
Trichloroacetic Acid (TCA) 5%, 10%
Sodium Chloride
Ice
Sand
Cheese cloth

Procedure
1. Weigh fresh sample and cut into small pieces
2. Grind the fresh sample in a cooled mortar with washed sand in 25mL of 10% TCA
3. Strain through cheese cloth
4. Rinse the mortar and pestle with a small amount of 5% TCA and strain the
washings through the same cheese cloth
5. Determine the volume of the TCA extract using a graduated cylinder
6. Transfer the extract into a 250 mL beaker, and with constant stirring, slowly add
two volumes 95% EtOH per volume of the TCA extract
7. Leave undisturbed until flocculation occurs
8. Keep the precipitate for the General test and Hydrolysis of Carbohydrates

PREPARATION OF PLANT STARCH

Materials:

Potato, Unripe Banana, Cassava or Corn
Cheese cloth
Filter paper
Mortar and pestle

Procedure
1. Peel and cut sample into very small pieces. Grind in a mortar
