LS23L Week 10 - Sequence Analysis and
Maternal Lineages Exam Questions and
Answers with Verified Solutions | Latest
Updated 2026

Lab Goal In this lab, you will receive your
mitochondrial DNA sequence, which was
sequenced off site after PCR
amplification. You will then use this
sequence to determine your haplotype
and create a student phylogeny tree, as
well as a class phylogeny tree.

Electropherogram / The data printout from Sanger
Chromatogram Sequencing. It shows peaks of colors,
with each peak corresponding to a
different nucleotide in the sequence.

RSRS Recent Sapiens Reference Sequence

Annotating your sequence Annotating your sequence means
attaching information to a sequence of
DNA. Go back to your 4-color data and
fill in all of our demographic information
to the best of your knowledge. Click on
the blast your sequence button, and then
hit search. This will bring up all of the
most likely haplogroups for you. Then hit
submit annotation.

, Creating Two Phylogenetic Click "make student tree" and then click
Trees Using Your Sequence on a letter upstream of your own
haplogroup. If you click on your group,
you will only see individuals with the
same haplogroup which is boring. A table
will appear with all the sequences in the
current data base belonging to the
haplogroups you selected. Click about 10
and then click Make Tree. Once you get
your tree, you will be able to look at each
sequence and see if any are too short. If
so, delete them. You can then run your
tree again. Continue until you are happy
with your tree.
Then, click Make Section Tree. Do the
same steps as you did for your student
tree.

The Recent Out of Africa Modern humans arose in Africa about
Hypothesis 100,000-200,000 years ago and spread
throughout the world by replacing the
local archaic human populations.

Neaderthals Left Africa about 300,000-400,000 years
ago (much before the modern human
left). However, after the modern human
left Africa, they overlapped for a bit (and
in fact, most people have about 1-4%
Neaderthal DNA.

Mitochondrial Eve The first group to split off when the rest
left Africa (stayed in East Africa?)
