The ‘Solver’ function in Excel was used to optimize the fit of the model to the data. Graph
1 shows the resulting dose-response curve for estradiol (E2).
Graph 1: The Dose-Response Curve for Estradiol (E2).
The x-axis shows the concentration of E2 in pM. The y-axis shows the Relative Fluorescence Units (RFU) in nm.
The parameters EC50 (median effect concentration), Fmin (minimum RFU), Fmax (maximum
RFU) and n (hill slope) were all read from the dose-response curve of estradiol (table 3).
Table 3: Parameters read from the dose-response curve of E2: Ec50, Fmin, Fmax and n.
EC50 655,75
Fmin 0,28
Fmax 0,99
n 1,67
The Analysis of Variance (ANOVA) indicated that the different concentrations of E2 had a
statistically significant difference in Relative Fluorescence Units (RFU), as shown in table 1
(colored yellow and marked with an asterisk).
