Ehrlich’s principle = zonder binding, geen effect

Het feit dat een drug werkt, geeft dat er een endogenous ligand is die aan dezelfde receptor bindt, en
tegenwerkt kan worden of gestimuleerd.

Moleculaire interacties
Binding vindt plaats, doordat het ligand in de receptor past, maar blijft door moleculaire interacties:
• Vanderwaals binding
• Ion binding
• Hydrogen
• Covalent (niet vaak bij drugs: irreversible)

What determines binding
Er is associatie & dissociatie gebaseerd op:
• Size
• Shape
• Charges

Stereospecificiteit door een assymetrisch C-atoom (= 4 verschillende groepen aan zich gebonden)
à bepaalt de shape à bepaalt of een ligand wel of niet past op een receptor

Types of drug targets
• Human cellulaire targets
o Proteins (receptor, enzym, ionkanaal, transporter, pomp)
o Nucleic acids
• Non human targets (vaak niet receptor-mediated)
o Chemical targets (ion, surfactant, bowel in GI tract)
o Micro-organisms
Drug target = any protein that can be reached with antibodies
