1. How do banding patterns change when a muscle contracts? the length of the band does not change during contraction of the muscles. However, the width of the banding pattern changes as the degree of overlap changes and results in shortening in the band. 2. What is the difference between a muscle organ, a muscle fiber, myofibril and a myofilament? Muscle fiber is a single muscle cell. Myofibrils are long and cylindrical that are found within the fiber. Myofilaments are the protein filaments and they make up the myofibrils which are responsible for the muscle contraction. 3. Outline the molecular mechanism for skeletal muscle contraction. At what point is ATP used and why? Occurs when myosin attaches to actin and pull them closer together, shortening the sarcomere. This process (driven by ATP) repeats as the muscle contracts and relaxes. Initiation, Crossbridge formation, power stroke, detachment and re-energizing, relaxation. 4. Explain why rigor mortis occurs. Rigor mortis is the stiffening of the skeletal muscles approximately 2 hours after death. This occurs due to the depletion of ATP, which is no longer being produced by the body. As ATP is no longer able to detach, muscles become locked in a contracted state, leading to stiffness and rigidity.

Pre- lab Questions

1. How do banding patterns change when a muscle contracts?
the length of the band does not change during contraction of the muscles. However, the width
of the banding pattern changes as the degree of overlap changes and results in shortening in
the band.

2. What is the difference between a muscle organ, a muscle fiber, myofibril and a myofilament?
Muscle fiber is a single muscle cell. Myofibrils are long and cylindrical that are found within
the fiber. Myofilaments are the protein filaments and they make up the myofibrils which are
responsible for the muscle contraction.

3. Outline the molecular mechanism for skeletal muscle contraction. At what point is ATP used
and why?
Occurs when myosin attaches to actin and pull them closer together, shortening the
sarcomere. This process (driven by ATP) repeats as the muscle contracts and relaxes. Initiation,
Crossbridge formation, power stroke, detachment and re-energizing, relaxation.
4. Explain why rigor mortis occurs.
Rigor mortis is the stiffening of the skeletal muscles approximately 2 hours after death. This
occurs due to the depletion of ATP, which is no longer being produced by the body. As ATP is no
longer able to detach, muscles become locked in a contracted state, leading to stiffness and
rigidity.

EXPERIMENT 1: TENDONS AND LIGAMENTS
Introduction Questions
1. How does the extracellular matrix of connective tissues contribute to its function?
Extracellular matrix is important as it provides structural support, facilitates
communication between cells and influences cell behavior.

2. Why are tendons and ligament tissues difficult to heal?
This is because both tendons and ligaments have limited blood supply, have a complex process
of tissue repair and the structure of their tissues.
Data and Observations
1. Label the arrows in the slide images below based on your observations from the experiment.
A. Chondrocytes
B. collagen
C. Collagen fibers
D. Skeletal Muscle fiber
E. Nuclei
F. Collagen Fiber
