1. Embryonic Origin of Head Structures
Mesenchyme Formation and Sources
• Mesenchymal Origins of head structures come from several sources:
o Paraxial Mesoderm:
§ Forms somites and somitomeres.
§ Contributes to:
§ The neurocranium (including both membranous and cartilaginous
components of the skull).
§ Voluntary muscles in the craniofacial region.
§ Dermis and connective tissue in the dorsal head region.
§ Meninges of the forebrain and midbrain.
o Lateral Plate Mesoderm:
§ Forms laryngeal cartilages (e.g., arytenoid and cricoid) and contributes to
connective tissue in the laryngeal region.
o Neural Crest Cells:
§ Originate from the neuroectoderm of the forebrain, midbrain, and
hindbrain.
§ Migrate into the pharyngeal arches and facial regions.
§ Form the viscerocranium (skeletal components of the face) and parts of the
neurocranium.
§ Also contribute to various structures such as cartilage, bone, dentin,
tendons, dermis, pia, arachnoid, sensory ganglia, and glandular
connective tissue.
o Ectodermal Placodes:
§ Work in coordination with neural crest cells to form neurons in the cranial
sensory ganglia (associated with cranial nerves V, VII, IX, X).
Pharyngeal Arches and Their Development
• Pharyngeal Arches (or branchial arches) are critical to the development of the head and
neck.
o They appear by the 4th and 5th weeks of development.
o Each arch is made up of a mesenchymal core, containing cells derived from both
mesoderm and neural crest cells.
o Pharyngeal Clefts separate each of these arches, and pharyngeal pouches form
along the lateral walls of the pharynx.
o Each arch contains its own:
, § Nerve, artery, muscle, and skeletal component.
• Pharyngeal Arch Organization:
o Each arch is a bar of mesenchymal tissue containing specific elements that
contribute to distinct parts of the face, neck, and surrounding structures.
o Arch mesenchyme is covered externally by ectoderm and lined internally by
endoderm.
2. Specific Contributions of Pharyngeal Arches
First Pharyngeal Arch (Mandibular Arch)
• Structural Components:
o Divided into two processes:
§ Maxillary Process: Gives rise to the maxilla, zygomatic bone, and part of
the temporal bone via intramembranous ossification.
§ Mandibular Process: Contains Meckel's cartilage, which forms the incus
and malleus (ear ossicles).
o Musculature:
§ Forms muscles such as temporalis, masseter, medial and lateral
pterygoids, mylohyoid, anterior belly of digastric, and tensor tympani.
o Innervation:
§ Supplied by the trigeminal nerve (CN V), specifically the mandibular
division (V3).
Second Pharyngeal Arch (Hyoid Arch)
• Cartilage Component:
o Forms Reichert’s Cartilage, which develops into:
§ The stapes (middle ear bone), styloid process, stylohyoid ligament, and
lesser horn and upper part of the hyoid bone.
• Musculature:
o Forms muscles of facial expression, including orbicularis oculi, orbicularis oris,
buccinator, stapedius, and stylohyoid.
• Innervation:
o Innervated by the facial nerve (CN VII).

• Cartilage:
o Forms the greater horn and lower part of the hyoid bone.
• Musculature:
o Forms the stylopharyngeus muscle.
