1.1. Signal Transduction is Essential for Survival and Omnipresent
in Biological Systems

 Cellulair niveau: een stimulus moet worden ontvangen en goed geïnterpreteerd worden
- Eencelligen: metabolisme en genexpressie aanpassen om te overleven
- multicellulair: verschillende celtypes moeten de juiste output leveren om een
fysiologische respons te coördineren
 Signaaltransductie: het proces waarbij een extracellulair signaal (chemisch, mechanisch of
elektrisch) wordt ontvangen, doorgegeven, geamplificeerd en omgezet in een cellulaire
respons

1.2. Signals are Transduced in Various Ways

1.3. Signal Transduction is Complex … but Contains a Simple
Blueprint

 Een stimulus kan het vrijkomen induceren of direct fungeren als een primary messenger
 Als de primaire boodschapper een molecuul is, wordt dit het ligand genoemd.
 Primaire boodschappers worden herkend door receptoren (meestal membraaneiwitten,
verschillende soorten)
 Ligand-receptor-interacties (cfr. begrippen specificiteit en affiniteit)
 Ontvangst van een primaire boodschapper door de receptor veroorzaakt een
conformatieverandering (cfr. allosterie)
 Liganden die aan de receptor binden en het effect van het natuurlijke ligand produceren
worden agonisten genoemd, terwijl liganden die de receptor binden zonder het normale
effect teweeg te brengen en die de effecten van agonisten blokkeren, heten antagonisten




2. The signal must be converted (transduction) and must be loud and clear (amplification)
 De primaire boodschap moet worden omgezet in andere chemische vormen (transductie)
 Conformatieverandering van de receptor leidt tot enzymactivering
 Enzymactivering zal uiteindelijk leiden tot de generatie van second messengers (cAMP,
cGMP, Ca2+, enz.)
 Amplificatie: een enkele geactiveerde receptor kan vele tweede boodschappers genereren,
die zich vrij in de cel verspreiden

 Voorkomen van scaffold proteins die nodig zijn om partners in elkaars nabijheid te brengen
 Signaalcascades zijn modulair
 Fosforylatie wordt vaak gebruikt als middel om het signaal door te geven; vaak voorkomen
van eiwitkinases
 Fosforylatie is omkeerbaar (belangrijk voor beëindigingsprocessen waarbij fosforylering door
eiwit fosfatases betrokken is)




 Eiwitten die werkzaam zijn in de signaalcascade (scaffold proteins en eiwitkinases) worden
signaaleiwitten genoemd
 signaaleiwitten zijn multivalent, d.w.z. zij kunnen met verschillende eiwitten tegelijk
