T – Zell – Rezeptor

- Ist ein Membranprotein
- Aus 2 Ketten: α – Kette und β – Kette
→ Heterodimer
- Eine konstante und eine variable Region
- Wird nicht sezerniert
- T – Zell – Rezeptor erkennt nur kleine Anteile des
Antigens
→ Einige, wenige Aminosäuren am Peptid
→ T – Zell – Rezeptor bindet Antigen nur, wenn diese von dem MHC – Protein
präsentiert werden




MHC (major histocompatibility complex)

- Klasse I = ist auf allen Zellen des Säugers zu finden (außer auf Erythrozyten)
- Klasse II = auf Antigen – präsentierenden Zellen (Makrophagen und B – Zellen)
- Bei Transplantationen von Geweben/Organen ist es wichtig, dass Spender und
Empfänger das gleiche MHC – Protein haben, damit es nicht zur Abstoßung kommt
- MHC bei Maus und HLA beim Menschen

- Besteht auch aus einer α – Kette und einer β – Kette
- Bindet mit seiner Bindungsstelle am N – Terminus das Antigen
→ Von der anderen Seite bindet dann T – Zell – Rezeptor

Antigenpräsentation

1) B – Zelle (reif, naiv)
→ Reif = fertiger Antigen – Rezeptor IgM auf der Oberfläche
→ Naiv = noch keine Massenproduktion Antikörper
→ Antikörper bindet Antigen
→ Antigen wird über Endocytose in die Zelle transportiert
→ Antigen im Endosom wird durch Lysosom zerlegt

2) MHC – II – Protein
→ MHC – II präsentiert ein Peptid – Fragment des Antigens auf der Zelloberseite

3) Effektor – T – Helferzelle
→ Passende T – Helferzelle bindet an Peptid – Fragment, welches an MHC gebunden
ist
→ Bildung einer immunologischen Synapse

4) Aktivierung B – Zelle
→ T – Zelle sendet Cytokine und CD40 – Liganden aus, die nur die gebundene B –
Zelle erreichen
→ Dadurch wird B – Zelle aktiviert

5) Aktive B – Zelle
→ B – Zelle differenziert sich zur Plasmazelle
→ Proliferation von Antikörpern
