 Thin layer silica gel chromatography plate
 Leaves, pestle and mortar

Chromatography is a method which is used to separate mixtures and to identify
substances. Through this method, we can find out the purity of a chemical substances.


METHODS FOR CHROMATOGRAPY EXPERIMENT


 Firstly, put the spinach into the bowl.
 Then, just cover the spinach leaves with
propanone, we should make sure that we
do not add too much of propanone.




 We should
grind these leaves thoroughly with the pestle until
we get smooth pulp.
 We then assessed the liquid that was made in the
bowl. If there is no liquid present, then we must
add little more propanone. If the liquid is not dark
green, then we must add more leaves and grind it
until we get a smooth paste.

 We then folded a filter paper and place it into the funnel
which was collected the filtrate in the test tube. We were
required a few drops of these filtrate.
 We must draw a line with the use of pencil across a thin
layer silica gel chromatography plate about 1 cm from
the end.




 By using micropipette,
we transferred pigment
extract to the TLC plate,
creating a small spot in the centre of the pencil line.


 We must allow the
spot on the TLC paper
to dry and then we
added another spot.
We repeated this,
until the spot was almost black, keeping the spot as
small as possible.


 We put the TLC plate in the chromatography tank in
the fume unit with the spot at the bottom. The solvent
level should not reach as far up as the spot itself.
 Then, we covered the tank with a black polythene bag
while the chromatogram runs. We lifted the black bag
every two minutes in order to check on progress of the solvent front.
 When the solvent front was about 2 mm from the above of the TLC plate, we
removed the chromatogram, we marked the position of the solvent front by using
pencil and left the fume unit for the solvent to evaporate. Make sure to replace the
lid on the chromatography tank.
 We then allowed the paper to dry.
 The paper was sprayed with ninhydrin by the teacher. Then, dry the paper using a
hair dryer in order to help the spots to develop.
