Hoofdstuk 1: Inleiding
De tricarboxyzuur (TCA) cyclus = De finale pathway waar de oxidatieve katabolisme van koolhydraten,
aminozuren en vetzuren samenkomen
-> De citroenzuur cyclus of de Krebscyclus
-> Koolstofskelet w omgezet tot CO2
-> Oxidatie biedt energie voor de productie van ATP
-> Gebeurt volledig in de mitochondriën
-> Dicht bij waar het elektronen transport gebeurt
Oxidatie van gereduceerde co-enzymen (NADH en FADH2)
-> Aerobe pathway: O2 nodig als finaal elektronen acceptor

Hoofdstuk 2: Reacties van de TCA cyclus
Er bindt eerst een acetyl groep van acetyl CoA op oxaloacetaat om de cyclus te starten
-> Oxaloacetaat w geregenereerd als de cyclus rondt is
-> Nood aan de inbreng van acetyl CoA
-> Leidt niet tot netto productie of consumptie van intermediaire
2.1 Oxidatieve decarboxylatie van pyruvaat
De hoofd bron van acetyl CoA is de oxidatieve decarboxylatie van pyruvaat
-> Pyruvaat is het eind product vd aerobe glycolyse
-> Pyruvaat moet vh cytosol getransporteerd w naar de matrix vd mitochondrium
Nood aan een specifieke transporter voor over het binnenste membraan te kunnen
Eens in de matrix vd mitochondrium w pyruvaat omgezet in acetyl CoA door het pyruvaat dehydrogenase
complex (PDH complex)
-> Dit is een multi-enzym complex
-> PDH complex is een proteïne ophoping van verschillende koppies van 3 enzymen:
Pyruvaat decarboxylase (E1), dihydorlipoyl transacetylase (E2), dihydrolipoyl dehydrogenase (E3)
-> Elk enzym katalyseert een deel van de reactie
-> Complex bevat ook 2 sterk gebonden regulatory enzymen:
Pyruvaat dehydrogenase kinase (PDH kinase) en pyruvaat dehydrogenase fosfatase (PDH fosfatase)
-> Het PDH complex bevat 5 co-enzymen die zich gedragen als carriers of oxidanten voor de
intermediaire van de reacties:
E1 heeft nood aan thiamine pyrofosfaat (TPP)
E2 heeft nood aan lipoic zuur en CoA
E3 heeft nood aan FAD en NAD+

REGULATIE VH PDH COMPLEX:
Covalente modificatie gebeurt door de 2 regulatory enzymen die deel zijn van het complex
-> Activeren en inactiveren E1
-> cAMP onafhankelijk
-> PDH kinase fosforyleert en inactiveert E1
-> PDH fosfatase defosforyleert en activeert E1
Het PDH kinase w geactiveerd door ATP, acetyl CoA en NADH
-> DUS wanneer de energie levels hoog zijn is het PDH complex geïnactiveerd
Pyruvaat is een krachtige inhibitor van PDH kinase
-> Als [pyruvaat] hoog is, is E1 maximaal
Ca2+ is een sterke activator van PDH fosfatase
-> Stimuleert E1 activiteit
-> Belangrijk is skelet spieren
-> Vrijlating van Ca2+ tijdens contractie leidt tot stimulatie PDH complex en zo energie productie
2.2 Synthese van citraat uit acetyl CoA en oxaloacetaat (OAA)
De condensatie van acetyl CoA en OAA vormt citraat
-> Aldol condensatie
-> Evenwicht ligt ver in de richting van citraat synthese
-> Mbv. Citraat synthase
-> Citraat synthase w geïnhibeert door zijn product (citraat)
-> Binding van OAA op het enzym veroorzaakt een conformatie verandering waardoor acetyl CoA ook
kan binden
OPM. Citraat inhibiteert ook fosfofructokinase 1 (PFK-1) -> glycolyse
Citraat activeert ook acetyl CoA carboxylase -> vetzuur synthese
2.3 Isomerisatie van citraat
Citraat w omgezet tot isocitraat
-> Door Aconitase (aconitaat hydratase)
