INLEIDING

Niet alle virussen zijn ziekmakende virussen:

• Heel groot deel zijn endogene retrovirussen:
o Grootste deel van ons erfelijk materiaal bestaat uit deze endogene retrovirussen. Deze
bepalen dus wie wij zijn.
o Voor een deel zijn deze niet meer actief, voor een deel weet men niet of deze nog actief
zijn.
• Virussen die ziekte makend zijn hebben niet de bedoeling om ziekte te verwekken/doden, ze
willen gewoon hun erfelijk materiaal vermenigvuldigen.

GESCHIENDENIS

Woord ‘virus’ eerste keer gebruikt op einde 19e eeuw door Martinus Bejerinck:

• Hij benoemt iets dat ultrafiltreerbaar is en ziekteverwekkend is bij tabaksplanten.
• Virus in het Latijn = vergif

Vanaf midden 20ste eeuw komt virologie in opmars door nieuwe technologieën zoals
elektronenmicroscopie, celcultuur,… opkomen. Het bereikt zijn piek in populariteit in de jaren 80.

Historisch belangrijke virussen:

• Pokken: ziekte is uitgeroeid dankzij vaccinatie.
• Polio: nog niet volledig uitgeroeid komt enkel nog voor tussen afghanistan en pakistan, polio-virus
infecteert zenuwbanen zodanig dat bepaalde spiergroepen niet meer kunnen werken.
o Als middenrif niet meer werkt moet men beademd worden, komen in ijzerenlong terecht.
• HIV/AIDS: in jaren 90 belangrijkste doodsoorzaak bij jonge mannen in de VS, door behandeling is
sterfte nu veel minder.

VIRUSSEN IN BEELD BRENGEN

Kenmerken:

• Zeer klein: 25- 400 nm
o Mimivirus = grootste virus dat er is (400 nm)
o Picornavirussen = kleinste virussen (25 nm), o.a. polio
• Ultrafiltreerbaar: filters die bacteriën tegenhouden laten virussen wel door
• Niet zichtbaar met gewone microscoop, elektronenmicroscoop voor nodig
o Met X-ray kristalografie kon men kristallen maken van virussen, zo kon men betere
resolutie krijgen om modellen te maken van virussen. Dit werd gedaan door Rosalind
Franklin.
o Op dit moment is er Cryo-electronmicroscopy, dit laat toe om met complete resolutie elk
atoom van de structuur van een virus te kunnen visualiseren (gekleurde beelden).
