H6 growth inhibition and tumor suppressor genes
6.1 Definitions of tumor suppressor genes
Tumor suppressor gene = inactivation of genes (both alleles) promotes
cancer
- Can be inherited when germline allele is mutated
- = knudsons two hit model / hypothesis
BRCA1 and BRCA2 = breast and ovarian cancer genes
BRCA2 recruits RAD51 and BRCA1 homologous recombination DNA
repair
PTEN = a lipid and protein phosphatase
- Converts PIP3 to PIP2 blocks the PI3K-Akt-mTOR pathway no
proliferation
- PTEN mutations cowden syndrome
PTPN1 = can act as tumor suppressor or and oncogene depending on
tissue
Some kinases, like ATM, can function as tumor suppressor
