LAB ACTIVITY 2: Calculating Magnification
1
Calculate the total magnification by multiplying the magnifying powers of your
microscope lenses.
• scanning lens __40__ × ocular lens _10__ = _40__ total magnification
• low-power lens _10__ × ocular lens _10__ = _100__ total magnification
BIOS251 Week v1 vLab vInstructions v(B) Page v1 vof v11
, • high-dry lens _40__ × ocular lens __10_ = 400__ total magnification
• oil immersion lens _10_ × ocular lens 100__ = 1000__ total magnification
• EXERCISE 3: Reviewing Your Knowledge
Name Section ____________________
•
• A. Care and Use of the Microscope
• Correct each statement by crossing out the incorrect word(s) and inserting the correct
word(s).
1. One hand is to be used to transport the microscope.
Two hands are to be used to transport the microscope, 1 under the scope the other holding it
the arm.
2. Tissue paper can be used to clean the microscope lenses and prepared slides.
Clean lens with optical safe lens special lens paper.
3. The microscope should be put away with the high-dry lens in position.
Microscope should be put away with less powerful lens position down.
4. The coarse focusing knob should be used when using the high-dry lens.
No never, This controls the stage so, it can cause the slide to crack .
5. The iris diaphragm should be completely open to obtain maximum contrast.
Depending on the specimen size the iris diaphragm can be open or smaller. The more open it is
the more light will pass threw
6. The condenser should be in the lowest position (far from the stage) to focus the
most light on the specimen.
Highest
B. Parts of the Microscope
Write the term that the phrase describes.
• _____Coarse focus knob____ 1.
Large knob that moves the stage or objective lens a great distance. Used with scanning
or low-power objective lenses only.
_____Stage____ 2.
Flat platform beneath the objective lens on which the microscope slide is placed.
____Ocular lens______ 3.
Removable lenses that you look through to observe the microscope slide.
____Fine Coarse_____ 4.
