Bij de gramkleuring wordt er onderscheid gemaakt tussen grampositieve en gramnegatieve bacterien. Het verschil berust op het verschil in celwand bouw. Gram-positieve bacteriën hebben een dikke peptidoglycaanlaag in de celwand die ondoordringbaar is voor ethanol waardoor de kleur niet wordt weggespoeld. De gramnegatieve bacteriën hebben een dunne celwand die de kleurstof niet vasthoudt.
Stappen:
1. 1 minuut in kristalviolet.
2. Spoelen met water
3. 1 minuut in lugol oplossing
4. Spoelen met 96% ethanol
5. 1 minuut ontkleuren met 96% ethanol
6. Spoelen met water
7. 1 minuut nakleuren met safranine
8. Afspoelen met water en laten drogen

De concentratie enzymen, co-enzymen als substraat zorgen ervoor dat de reacties sneller of trager verloopt.
