bacteriële celwand.
Kristalviolet is positief
geladen en zal dus hechten
aan negatief geladen
structuren in de
peptidogycaanlaag zoals de

Ethanol Zorgt voor het wegspoelen
van de lipidenlaag rond
Gram-negatieve bacteriën
waardoor het kristalviolet-
jodiumcomplex wordt
weggewassen.
Fuschine Kleuren van bacteriecellen.
Het is positief geladen en
bindt dus aan negatief
geladen structuren in de
peptidogycaanlaag zoals de
Teichoic acid en
lipopolysachariden
Jodium Vormt een complex met
kristalviolet door middel
van elektrostatische
interactie, hierdoor wordt
het kristalviolet gefixeerd
in de celwand.
KATALASE ASSAY
(3%) Waterstofperoxide Wordt gebruikt om te
(H2O2) kijken of er katalase-
activiteit is. Als hier sprake
van is zal het namelijk
omgezet worden tot water
en zuurstof waarna er
gasbelvorming ontstaat.

wordt met UV-licht. Op
deze manier is het mogelijk
om de 16S rRNA-bandjes
in de gel te visualiseren en
dus om te controleren of je
PCR gelukt is.
