Title: 01 Introduction to Medical Microbiology



Objective:

Cultivation of lab samples (growth conditions) that are necessary for carrying out experiments
and study pathogens in a lab setting.

Procedure:

• Equipment Used:

o Keep a sterile environment to keep people healthy

o The tool for this is the Autoclave.

▪ Cleans equipment at 125⁰C

o Growing: Fixed incubator or a shaker incubator

▪ Fixed incubator operates at 37 C

▪ Storage: A refrigerator at 4 C

o General Lab Safety:

▪ Never eat or drink in the lab

• Contamination risks

▪ Use personal protective equipment (PPE)

Equipment Used:

o Keep a sterile environment to keep people healthy

o The tool for this is the Autoclave.

▪ Cleans equipment at 125⁰C

▪ Uses heat pressure and steam to sterilize your environment.

▪ A chamber with a thick door that will be closed with equipment or a
sample inside to sterilize.

▪ Uses hot steam instead of hot air because it sterilizes in a matter of
minutes verses several hours.

▪ When opening the chamber after a cycle is complete, be sure to open
slowly and carefully after it has depressurized to avoid being burned by
the hot steam that will be released.

o Growing: Fixed incubator or a shaker incubator

▪ Fixed incubator operates at 37 C that houses petri dishes, which holds
samples, and tubes which hold liquid cultures. It has a door that seals
such to maintain dark conditions for growing.

▪ Shaker: growth media, a liquid nutrient broth, is placed in the metal
holders to be shook. The shaker is used to inoculate bacteria as it rotates
the sample to aerate it and provide optimal conditions for bacteria
growth.

▪ Visualization: Microscopy

▪ Storage: A refrigerator at 4O C is used to slow the growth of bacteria and
preserve samples. Petri dishes are wrapped in a para film, a waxy film to
keep it airtight.

o General Lab Safety:

▪ Use personal protective equipment (PPE)

• non-latex available usually purple, blue, or green
