BIOL 2053 FINAL EXAM COMPLETE PRACTICE
EXAM QUESTIONS AND ANSWERS NEW UPDATE
Louisiana State University
Final Exam (Grade 1) for Principles of Genetics, Fall Term, Section 5-10. Bubble in
your name and student identification number on SIDE TWO of the scantron. Also, you
need to mark in the grade section whether your exam is Grade 1 or 2. The correct
answer for each question will be awarded 3 points (3 x 34 = 102). Keep this exam
booklet for your record.

(Q1-4) In fly, one unusual phenotype has been discovered from a strain called A,
which has an extra pair of legs. According to the breeding results, this mutant allele
with 8 legs termed n is recessive to the normal allele with 6 legs termed N. We
performed linkage analyses with three individual SSLP markers: SSLP#1 from
chromosome 1, SSLP#2 from chromosome 2, and SSLP#3 from chromosome 3. All
of these SSLP markers generate 200 bp products from Lab strain, while each of
these markers derives 250 bp products from the A strain. This linkage analysis
involved the following set of breeding experiments: the first crossing two pure-bred
Lab and A strains and the second crossing F1 and A strains. The subsequent F2
progenies were genotyped at 4 loci, which include the mutant locus, SSLP#1,
SSLP#2, and SSLP#3.

Lab (N/N; SSLP#1,2,3, 200/200) x A strain (n/n; SSLP#1,2,3 250/250)

F1 (N/n; SSLP#1,2,3, 200/250) x A strain (n/n; SSLP#1,2,3 250/250)

The following represents the summary of genotyping results from 100 F2 progenies:

F2 category SSLP#1 SSLP#2 SSLP#3

Cat1 N, 250 26 25 11

Cat2 n, 200 24 25 9

Cat3 n, 250 25 26 41
Cat4 N, 200 25 24 39

B. SSLP#1 and SSLP#3
C. SSLP#2 and SSLP#3
D. all of the above
E. none of the above

2. This mutant allele is located on which chromosome?

, 3. Which ones represent the recombinant types among the 4 categories?
A. Cat1 and Cat2
B. Cat3 and Cat4
C. Cat1 and Cat3
D. Cat2 and Cat4
E. Cat1 and Cat4

4. What is the mapping distance between the mutant locus and the linked SSLP?
A. 4%
B. 8%
C. 12%
D. 16%
E. 20%

(Q5-9) The picture shown below illustrates how we can genotype sickle cell anemia with
molecular biology techniques. Given the information we learned through the lectures,
answer the following questions.

,5. Which technique was NOT used for generating the results shown in (b)?
A. PCR
B. Restriction enzyme digestion
C. Gel electrophoresis
D. Southern blotting
E. Pedigree analysis

6. The 'Left Primer' will NOT be part of which product?
A. 500 bp from S allele
B. 500 bp from A allele
C. 300 bp from A allele
D. 200 bp from A allele
E. all of the above

7. In the pedigree (b), what is the genotype of the daughter?
A. A/S
B. S/A
C. A/A
D. S/S
E. none of the above

8. Which statement is INCORRECT about the sickle cell anemia of this family?
A. transversion mutation
B. mis-sense mutation
C. gain-of-function mutation
D. dominant trait
E. haploinsufficiency

9. If this family has a vacation to a Colorado mountain, which member will NOT have
any dizziness?
A. Father
B. Mother
C. Son
D. Daughter
E. all of the above

, (Q10-12) The diagram shown below illustrates the genotyping results of one family
with the Huntington's disease. Both parents and the first child are affected with
this disease, but the second child is normal without this disease. Also, Allele 2 turns
out to co-segregate with this disease. While performing this genotyping,
unfortunately, we made two major mistakes: 1) we did not label the PCR reaction
tubes, and also 2) we accidently included two additional samples that are not
related to this family. Thus, we do not know which genotype represents the allele
combination of each family member. The following shows the PCR results from 4
family members plus two unrelated samples.

10. Which allele combinations represent the genotypes of two parents?
A. 1/1 and 2/2
B. 1/1 and 3/3
C. 1/2 and 2/3
D. 2/2 and 1/3
E. 1/3 and 2/3

11. Which allele combinations represent the genotypes of the first and second
children, respectively?
A. 1/1 and 2/2
B. 1/1 and 3/3
C. 1/2 and 2/3
D. 2/2 and 1/3
E. 1/3 and 2/3

12. Which allele combinations represent the genotypes of the two unrelated
samples?
A. 1/1 and 2/2
B. 1/1 and 3/3
C. 1/2 and 2/3
D. 2/2 and 1/3
E. 1/3 and 2/3
