Starting from your problem description, explain the goal of your project in your own words. Will opt
for a forward or reverse genetics approach? Provide an argumentation.

The goal is to identify the genes that contribute to the process how light is detected by
photoreceptors in the eye. Because in this case it is about identifying genes I choose to
use forward genetics.
Forward genetics is the approach of determine the genes responsible for a phenotype.
This is done by using mutations with radiation, chemicals or transposons.

Choice of model organism (e.g. 8-12 sentences)
Indicate for each of the four model organisms (mouse, fly, worm, yeast) if they would be suitable.
Which model organism will you choose for your project? Motivate your choice, also considering the
evolutionary conservation if you have a predefined gene of interest.

Yeast, they are not suitable because they don’t have eyes. The eyes are very essential
in this research.
Mouse, generation time is long. Genome is big. So, not that suitable because you need
a lot of mouse and this takes very long.
Fly, Generation time is 10 days, genome is small. Not that suitable because generation
time is long.
Worm, they are transparent organisms because of detecting light this may be useful.
Also, there generation time is short only 3 days and they can self-fertilize. Worms don’t
have eyes but they can detect light so they have photoreceptors.

So, the worm will be the model organism in the research. Because it is a transparent
organism and because in this experiment light is detected this may be useful they also
have photoreceptors. Beside that the generation time is very short and they are able to
self-fertilize, these two are advantages for forward genetics.

Choice of technique (e.g. 6 – 10 sentences)
Indicate for each technique (transgenesis, knockout/knockin, conditional knockout/knockin, RNA
interference or random mutagenesis) if it could be applied for your purpose. Why/why not? What is
your final choice?

The only choice for forward genetics is the technique random mutagenesis.
