Pre-Lab Questions
1. Of the four major types of microscopes, give an example of a scenario in which each
would be the ideal choice for visualizing a sample.
• Stereo (Dissecting): Examination of small macro organisms or structures too large
to be visualized with a compound microscope. Used in teaching and research labs.
• Compound: Inexpensive way to look at microscopic cells and other features. Used
in teaching and research labs.
• Confocal: Can be used to visualize individual components. Used in medical and
research labs.
• Electron: Can be used to examine the most minute structures. Used in large
medical and research labs.

, 2. What is the difference between the coarse adjustment knob and the fine adjustment knob?
When is it appropriate to use each of them?
Coarse adjustment knob allows for large adjustment of the stage where fine adjustment
knob allows for precise adjustment.
3. If you are using a compound microscope with an ocular lens with a magnification of 10x
and an objective lens with a magnification of 40x, what is the total magnification?
400x
4. In this lab, we discussed preparation of a wet mount slide. Research and describe another
slide preparation and an example of when it would be used.
Dry mount slide: Put a few drops of liquid in the concavity and add water to thin it out if
necessary. A dry mount slide can be prepared in a very similar way to wet mounts,
except without the water. Place the object in the middle of the slide, and holding the
cover slip at an angle, carefully set it on top.
