Mitose→ Niet de complete celdeling (1 tot 2 uur van de 20-24 uur dat de humane celcyclus duurt)
Oöcyt = Eicel
• Ideaal voor celcyclus onderzoek
o Mooie grootte (~1mm)
o Makkelijk te injecteren
o Bijgedragen aan onderzoek naar celcyclus
o Bevindt zich standaard in de G2 fase tot deze bevrucht wordt (Gesynchroniseerde
celpopulatie = Alles bevindt zich in de G2, tot het bevrucht wordt.)
• Bij bevruchting gaat deze eerst heel snel door de M en de S fase zonder groei → Wordt eerst
dus kleiner, maar groter aantal cellen

Celcyclus in zoogdiercellen
Onderstaande afbeelding geeft relatief goed de verhouding in tijd weer per fase
G1 fase → ‘Gap’ fase 1 - Diploïd (1 set DNA)
S fase → DNA-replicatie - Verdubbeling DNA
G2 fase → ‘Gap’ fase 2 - Tetraploïd (2 sets DNA)
M fase → Mitose (kerndeling) en Cytokinese (cytoplasmatische deling) - Tetraploïd (2 sets DNA)
Experimenteel kan de fase bepaald worden:
Piek A -> Diploïd
Dal S -> Replicatie DNA
Piek B -> Tetraploïd
Hoe weten we hoe de celcyclus verloopt?
• Eiwitten in de celcyclus zijn sterk geconserveerd in
eukaryoten
• Gebruikte eiwitten in eukaryoten zijn erg
vergelijkbaar met elkaar
o Dusdanig dat humane eiwitten gebruikt
kunnen worden in gist
→ Injectie kan oöcyt de
celcyclus laten vervolgen
→ Daaruit blijkt dat er dus
iets in het cytoplasma zit
van cellen in de M-fase die
celdeling/vervolg celcyclus
aan kan zetten

Datgene wat de cel van G2-fase naar M-fase over laat gaan: MPF
• Maturation Promoting Factor is een eiwitcomplex (MPF)
o Bestaande uit een cycline en een CDK
• Laat een cel naar de volgende fase gaan
Groen = Actief MPF
Neemt snel toe en snel af
rond de mitose
Cel fusie experimenten
Er is dus specificiteit & er zitten factoren in het cytoplasma die overgang naar de volgende fase
mogelijk maken. Dit gaat dus niet ‘uit zichzelf’.
