Human life depends on the process of cell division. Important for cell growth,
repair and regeneration. The type of cell division that takes place depends on
the type of dividing organism. Eukaryotes divide cells using mitosis and meiosis
in sexual reproduction
Preparing a microscopic slide:
The purpose of this task is to teach you how to prepare and dye a root tip so you
can view the stages of mitosis under a microscope.
Appratus:
Garlic roots
Razor
1moldm¯³ of hydrochloric acid
Acetic orcein
100ml beaker
2 watch glasses
Dropping pipette
Microscope slides and cover
Filter paper
Microscope with x100 & x400 magnification

Method:
• Prune a lateral root that is 5 mm in length from the tip.
• To prepare the root tip, combine it with 20 drops of acetic acid and 2 drops
of 1 millilitre HCI in a watch glass.
, • Place the watch glass on top of a 100ml beaker of boiling water and heat
it for 5 minutes. By now, the acid will have disintegrated the tissue.
A clean microscope slide should be placed over the dyed root tip. The root
should be cut in half transversely, with the portion furthest from the apex
being removed. Maintain the pointed tip, please.
• Place two or three drops of acetic acid on the slide's root tip.
• Use the flat end of a stirring stick to compress the root's tip.
