Benzodiazepines- Summary
Mechanism: Bind to GABAA channel & increase
frequency of channel opening & increase chloride ion
conductance and cause CNS depression(GABA
facilitatory)- do not open channel directly.
Site – Midbrain, ARAS and limbic system.
Prototype drug- Diazepam
Long Acting – Nitrazepam, Diazepam, Lorazepam
Short acting – Alprazolam, Triazolam
Actions-
CNS – Sedation, anterograde amnesia, anxiolytic,
muscle relaxant, anticonvulsant
Also reduce nocturnal gastric acid secretion
Uses
1. Insomnia
2. Anxiety
3. Epilepsy – status epilepticus & febrile seizures
4. Preanaesthetic medication
5. Muscle Relaxant
6. Alcohol withdrawal
7. IV Anaesthesia ( Midazolam)
