The main purpose of the objective section is to keep the lab notebook organized and conveniently have access to a summary of completed work for each lab module when looking back on the notes. For the purpose of lab 1, this objective is to cover exactly how to maintain a proper, thorough, and well-organized lab notebook. Keeping a well-organized lab notebook is very important and will be very useful. The lab notebook will serve as a template to keep important details regarding experimental records, procedures, notes and results. Procedure: The procedure section of the lab notebook will be utilized to list out the steps for each lab 1. Not only will each step a given protocol be recorded in the procedure… i.e. what kind of chemicals, temperature, growth conditions, plates, etc. 2. But also, any deviations that may occur during the experiment will be clearly indicated in the procedure anything that differs from standard protocol which may result in different experimental outcomes) Notes: Any additional information will be included in the notes section that of which is helpful but is not listed in the procedure 1. Course Objectives: Cultivation of samples (growth conditions) equipment used in lab settings Identification of samples (biochemical assays) tests available look and determine what kind of bacteria/pathogen a patient may have Evaluation of samples (microscopy) visualization and recognition of key characteristics of each of pathogen 2. Basic Equipment: Cleaning – maintaining a clean and sterile environment is essential for our health and safety Autoclave: a tool using heat, pressure, and steam to sterilize lab equipment before or after use - 125o C - uses hot steam, which is more efficient than dry hot air - to prevent injury from hot steam, one must take caution when opening by: ensuring it is depressurized, opening slowly, wearing thermal gloves Growing – the bacteria/pathogens we observe Fixed incubator: chamber with an airtight metal door to seal and keep conditions dark - 37o C Shaker incubator: used to take growth media (liquid nutrient broth) to inoculate bacteria - it shakes and rotates culture in a swirling fashion to help aerate it and optimally grow - 37o C Visualizing Microscop
