• Cells are broken up and organelles are separated
• Before fractionation, cells are in a cold, buffered, isotonic solution

• Cold – reduce enzyme activity so organelles are whole
• Buffered – stops pH changes
• Isotonic – prevent bursting/shrinking due to osmosis
