The superoxide dismutase study (A. Janssen et al. (1998), data modified) concerns the role of
the oxidant-antioxidant balance in the initiation, promotion, and therapy resistance of cancer.
Superoxide dismutase (SOD) is a key antioxidant enzyme, scavenging the superoxide radical
(O2 –), which is a precursor molecule for all other reactive oxygen species and their
derivatives. By scavenging O2 –, SOD form the first line of defence against oxidative stress
and its subsequent effects. In human cells three isoenzymes of SOD exist: cytosolic (Cu,Zn-
SOD; SOD1), mitochondrial (Mn-SOD; SOD2) and extracellular (EC-SOD, SOD3).
Disturbances in the functioning of the SOD isoforms lead to numerous pathological changes
in the human organism, including tumor disease. In order to get more insight into the
relationship between SOD expression and the development of colorectal cancer the SOD
concentration (ng/mg protein) of normal mucosa and carcinoma tissues of colorectal cancer
patients (n=271) was measured and related to several clinicopathological parameters and the
survival time of the patients.
Relevant variables:
pat_number: patient number
msode_normal_mucosa: Mn-SOD in ng per mg protein of normal mucosa
msode_carcinoma: Mn-SOD in ng per mg protein of carcinoma
surviv: survival, coded 0 for survivors, and 1 for non-survivors.
survt: survival time in months
survt_grp: survival time classified into 4 categories, 1= 0.5-10 months; 2=11-57 months; 3=58-
105 months; 4=106-157 months
tnm01: developmental stage of carcinoma, 0= early development stage (TNM1 & 2; 1 advanced
development stage (TNM3 & 4). TNM (Tumor, Node, Metastasis) is an international accepted
staging system for colorectal carcinomas ranking from 1 to 4.
gender01: gender of patient, 0= female; 1=male
a. Perform a one-sample t-test to test whether the average Mn-SOD in carcinoma tissue
at population level is equal to 800 ng/mg protein. Perform the analysis using the
following steps
