Graticule
- Graticule is small disc that has an engraved ruler-
on the eyepiece of the microscope
- Does not move when you move specimen
- Size does not change when you change the
objective lens
- One division of the eyepiece represents a
different length depending on magnification
Stage Micrometre
- Slide with a scale marked on it
- Placed on stage of a microscope
- Usually 1mm long and marked in hundredths
- Used to find out how many micrometres each
graticule unit represents
Calibrating:
- Specimen removed from stage and replaced with
stage micrometre
- Use scales together to work our each graticule
unit
- Graticule then used as a ruler in the field of view
Method:
- Place a stage micrometer on the stage of the
microscope.
- Line up one of the divisions on the eyepiece
graticule with a fixed point on the stage
micrometer.
- Count the number of divisions on the eyepiece
graticule that correspond with a set
