Urine: Uitwendige lijkschouwing

- Aanwezigheid van metabolieten wijst op een vroeger gebruik
- Hogere concentraties dan in bloed
- Geneesmiddel of drug langer aantoonbaar.

i. Toepasbaar op meerdere matrices
ii. Staal opzuivering, extractie van gewenste analyten
(geneesmiddelen)
2) Dunne-laag chromatografie Toepasbaar op meerdere matrices
i. Scheiding o.b.v. polariteit (retentie)
3) Identificatie
i. Visualisatie o.b.v. UV-licht
ii. Vergelijking met referentiestandaarden



 Vloeistof extractie


Principe staalopzuivering:

Vloeistof-vloeistof extractie houdt het gebruik van 2 vloeistoffen in: -
organisch
- waterig

Verdeling van analyt over beide fasen o.b.v pH (ionisatie):

- Geïoniseerd = voornamelijk aanwezig in waterige fase
- Neutraal = overgang naar organische fase tot bereik van evenwicht

Doel: geneesmiddelen extraheren in organische fase

- Kan worden drooggedampt + gereconstitueerd
- Aanconcentrering van staal (daardoor gemakkelijker te visualiseren)

Hoe?

- pH aanpassen van waterige fase
- fasen mengen om overgang van neutrale componenten te
bewerkstelligen
