Principe:

Glucose-gehalte bepalen in plasma via de hexokinasemethode

 Plasma is complexe matrix
 Specificiteit nodig zodat methode enkel werkt op glucose, niet
andere componenten in matrix
 Doe dit via colorimetrische methoden


Colorimetrische methode (= Somogyi–Nelson methode):

- Glucose is een reducerend suiker
- Glucose + Cu2+ --- reduceren ---> Cu+
- Arsenomolybdaat + Cu+ -----> blauwe kleur, meetbaar met
colorimetrie
- Hoeveelheid kleur meten met spectrofotometer linken aan
concentratie glucose dat initieel aanwezig was

!! beperking colorimetrische methode: in plasma ook andere
reducerende suikers

 Reageren ook met koper-reagens
 Overschatting van glucosegehalte = niet specifiek genoeg



Meest specifieke methode, enzymatisch

 Glucose specifieke enzyme = hexokinase
 Hexokinase: glucose + ATP ---- fosforylatie -----> Glucose-6-
fosfaat + ADP

!! glucose-6-fosfaat is ook foto-inactief (= zoals glucose)

,  Tweede enzym-reactie invoeren = glucose-6-fosfaat
dehydrogenasen
 G6PDH: Glucose-6-P + NADP+ ---oxidatie---> 6-fosfogluconaat +
NADPH + H+

!! NADPH is foto-actief die fotospectrometrisch gekwantificeerd
kan worden

- Glucose-colorimetrische methode: Somogyi-Nelson methode (PL1)
