Inoculation - the implantation of microorganisms into or upon culture media
Inspection - The colonies of broth cultures are observed microscopically for growth
characteristics (color, texture, size) that could be useful in analyzing the specimen content
Total magnification - the objective lens x ocular lens
Numerical aperture - a mathematical constant that describes the relative efficiency of a
lens in bending light rays
Culture - the observable growth that later appears on a medium for study
Axenic culture - a culture that is free of other living things, except for the
microorganism being studied. AKA pure culture
Sub-culturing - a new cell or microbiological (pure) culture made by transferring some
or all cells from a previous culture to fresh growth medium
Compound microscope - a microscope with two or more lenses.
Condenser lense - focuses light onto the specimen at one single point
Resolving power - A measure of the clarity of an image; the ability of an optical
instrument to show two objects as separate.
Bright field microscopy - Shines light through sample and magnifies the image with a
series of lenses; used on stained, live specimen
Dark field microscopy - use to visualize living organisms that would be distorted by
drying or heat or that can't be stained; light goes on the sides of the specimen
Phase contrast microscope - used to see dense areas of specimen
Florescent microscope - type of light microscope that uses an ultraviolet light; useful to
diagnosing infections
Scanning confocal microscope - uses a laser beam of light to scan the specimen
integrates images to allow focus on multiple depths or planes
