2. De Nucleus

= De plaats waar de info over alle celstructuren en activiteiten gecodeerd
ligt in het DNA van de chromosomen

 Functies:
- Geen eiwitproductie, enkel eiwitimport
- DNA replicatie: synthese + verwerking van 3 types RNA: rRNA,
mRNA, tRNA
 Vorm: Meestal centrale ligging, rond of uitgerokken => vorm van de
kern verraadt vorm van de cel
 Componenten:
- Nucleaire enveloppe (= dubbele membraan)
- Chromatine
- Nucleolus: hier zit de info om ribosomaal RNA te synthetiseren
(rRNA)
- Nucleaire matrix = cytoplasma -> cytosol




Donkere plekken = nucleoli




De nucleaire enveloppe

= Twee membranen gescheiden door nauwe ruimte (de perinucleaire
cisterna)

 Kenmerken:
- In contact met het RER

, - Fibreuze lamina aan de binnenzijde
(lamine A, B, C)
- Chromosomen in contact met fibreuze
lamina bij niet-delende cellen
- Nucleaire poriën: fusie van beide
membranen met een 8-hoekig eiwit
 Zorgen voor het transport:
Ionen en kleine diameter hebben vrije doorgang
Grote diameter via receptors en ATP-verbruik




Chromatine

= chromosomen in een niet sterk gecondenseerde vorm

- Heterochromatine (HC) vs. Euchromatine (EC)
Hetero = sterker gewonden, euchromatine = minder gewonden
- Histonen: gewonden DNA-streng rond basisch eiwit
- Basisstructuur = nucleosoom
- Ruimtelijke ordening ifv condensatie tot meiose of mitose-
chromosoom
- Veel euchromatine (licht gekleurde kern) => meer DNA
oppervlakte bereikbaar => hogere transcriptie-activiteit




Nucleolus

= Bolvormige structuur, rijk aan eiwitten en rRNA

 Regio’s:
- Nucleolair organizer DNA: basen die coderen voor rRNA
- Pars fibrosa: primary transcripts van rRNA genen
- Pars granulosa: rijpende ribosomen, inbouw van eiwitten
(ribosomen worden gevormd in de nucleolus)
 Functie:
- Kleine en grote ribosomale subeenheden worden gevormd en
verlaten de kern via de nucleaire poriën
