A. Identification
• Motility test -> use a wet mount procedure!
○ Live organisms are placed on coverslip, covered in water, and observed.
○ VERY different from Gram stain procedure in that organisms observed in a
Gram stain are dead and fixed.
○ Group movement is force of coverslip pressure, NOT indicative of motility!
○ -> Individual movement is key!
○ Since there is no stain, the slide itself will be marked. Can focus cells and
water in that area.
• Endospore test
○ Green stain is used to identify spores. Safranin-O is used as counterstain. -
> Green spores in pink cells!
○ While performing the spore stain you forget to heat the first step. What do
you expect to see? The spore w/o color and the rest of the cell pink.
Heating is used to soften the spore's protein coat and allow the malachite
green to enter the spore; w/o heating, the dye cannot penetrate the spore in
the first place.
