Samenvatting lesstof genetica periode 2.1Les 1: DNA en DNA isolatie
Van DNA naar RNA is transcriptie, andersom is reverse transcriptie. Van RNA naar eiwit is
translatie. Tussen transcriptie en translatie vind RNA editing plaats.
RNA editing: van pre-mRNA naar mRNA intronen verwijderen, exonen aan elkaar splicen,
poly-A tail, 5’ cap.
UTR= untranslated region. Bevind zich voor en na de start van het coderende deel (dit blijft er
dus na RNA editing. na de UTRs bevinden zich de 5’ cap en de Poly-A-tail.)
Dna bestaan uit 2 anti parallelle, alpha helische, niet-covalent gebonden complementaire
strengen gemaakt van dNTP’s: deoxyribosenucleotrifosfaten.
Purines: Guanine adenine . Een 6 ring en een 5 ring.
Pirimidines: Thymine (uracil) cytosine. Alleen een 6 ring.
G-C = 3 Hbruggen
A-t = 2 Hbruggen
Nucleotide = stikstofbase + suiker + fosfaat
Nucleoside = stikstofbase + suiker
De fosfaatgroep bindt zich aan de OH groep op het 3e koolstofatoom van het suiker onder
afsplitsing van water. Hierdoor ontstaan een fosfodiester binding.
Verschillen tussen RNA en DNA: e.s. vs d.s., dUTP (uracil) vs dTTP (thymine), ribose vs
deoxyribose, minder stabiel, verlaat kern.
Ribose heeft een OH groep op het 2e koolstofatoom, deoxyribose alleen een H.
De major groove is de grote “golfbeweging in de DNA helix . De minor groove de kleine
Bij transcriptie: 3’ 5’ is template. 5’ 3’ is “gelijk” aan 5’3’ mRNA, dit heet de coderende streng.
Daarna weer aflezen m.b.v. tabel voor eiwitten. AUG is start. Stop UAA UAG UGA.
Reading frames: hierbij kunnen van 1 streng 6 verschillende reading frames ontstaan door aan
beide kanten van de strengen 3 keer een base te verschuiven.
