Alkaptonurie:
Mendeliaans, autosomaal recessief overerfbare aandoening. Door een gendefect op
chromosoom 3 is er een defect in het homogenisaat 1, 2-dioxygenase. Hierdoor is er een
opstapeling van homogenisaat, wat leidt tot zwarte urine, gewrichtsproblemen, problemen
met hartkleppen, arthrose en ochronose: donkere verkleuring van oren, oogwit, …
G6PD-deficiëntie
De stofwisseling van glucose-6-fosfaat is niet goed in rode bloedcellen; wat leidt tot
hemolytische crisis, icterus, … Maar biedt wel resistentie tegen malaria.
Congenitale Glycosylatie Ziekten
Slecht geglycosyleerde eiwitten.
Hypercholesterolemie
LDL partikel stapelt op door een afwijking in de LDL receptor.
Gangliosidosen
Gangliosidosen stapelen op, leidt tot mentale aftakeling.
Fenylketonurie
Is een autosomaal recessieve aandoening, waarbij er een probleem (mutatie in het
PAH-gen) is in het omzetten van fenylalanine in tyrosine: resulteert in minder tyrosine.
Fenylalanine wordt omgezet naar fenylketonen en verwijderd via de urine. Tyrosine, waar
een tekort van is, is een voorloper van neurotransmitters (dopamine en noradrenaline), leidt
tot psychomotorische achterstand, kan verholpen worden met een bepaald dieet. Tyrosine is
ook een voorloper van melanine, te weinig melanine leidt tot hypopigmentatie,
visusstoornissen en fotosensitiviteit. Door de te grote hoeveelheid fenylalanine: is er een
competitie tussen andere aromatische aminozuren: is nadelig voor tyrosine en tryptofaan.
Behandeling via dieet: weinig eiwitten eten, en zo weinig mogelijk fenylalanine, weinig
inname van tyrosine: als je dit onmiddellijk doet na de geboorte is er een goede ontwikkeling
van het centraal zenuwstelsel, maar je moet dit je hele leven doen en vraagt dus veel inzet.
Glycogenose
Probleem met afbraak van glycogeen: leidt tot hypoglycemie, spierzwakte, hartfalen.
Albinisme
Omzetting van tyrosine in melanine gaat fout: minder melanine: lichte huid door afwezigheid
van pigment.
Tyrosinemie type 1
Door een afwijking in het FAH-gen is er meer tyrosine in het bloed, dat niet wordt afgebroken
door een defect enzym: de lever takelt af: wat leidt tot levercirrose, leverfalen of zelfs
levertumor.
