Functionele organisatie van de cel
Bespreek de asymmetrie in de lipide compositie van de celmembraan: hoe wordt dit gegenereerd en
wat zijn de implicaties. Wat is de functie van cholesterol?

Celmembraan
Cel is omhuld door de plasma of celmembraan dat een fostolipide dubbellaag (bilayer met een
buitenste en binnenste leaflet) is en het intracellulaire milieu scheidt van het extracellulaire milieu

Fosfolipiden:

▪ Hydrofiele kop
▪ Hydrofobe staart
 Fosfolipiden zijn amfipathisch
 Fosfolipiden hun vetzuurstaart is gekoppeld aan een glycerol (vandaar naam triglyceride)
 Sfingolipiden hebben een sfingoide (ipv glycerol bij fosfolipide)

Er zijn verschillende soorten fosfolipiden:

▪ Hydrofiele kop kan verschillen
▪ Lengte vetzuurstaart verschilt
▪ Verzadigdheid vetzuurstaarten:
▪ Verzadigd: geen dubbele binding (recht)
▪ Onverzadigd: dubbele binding (knik)

Receptoren in membraan moeten van conformatie kunnen veranderen, dit kan enkel als de
membraan een soort van vloeibaar is

Vloeibaarheid van het membraan is temperatuursgevoelig (Tm = transitie temperatuur tussen gel-sol
state):

1) Bij transitietemperatuur:
Overgang van geordende vaste toestand nr ongeordernde vloeibare toestand
2) Bij toevoeging cholesterol:
Overgang van ongeordende toestand nr geordende toestand
Bij te veel cholesterol is de membraan TE vloeibaar
3) Bij toevoeging onverzadigde vetzuren:
Overgang van geordende toestand nr ongeordende toestand (door de knik zal de
interactie minder goed zijn)
4) Hoe korter vetzuurstaart, hoe minder interactie, zwakke pakking, hoe vloeibaarder

Plasmamembraan: geordend, vast, solid <-> ongeordend, vloeibaar, liquid

Zuiver lipiden membraan:

▪ Staart kort & onverzadigd → lage Tm → makkelijker om van gel-sol nr liquid
▪ Staart lang & verzadigd → hoge Tm → moeilijker om van gel-sol nr liquid
