Introduction to Microscopy



Objective:

To become familiar with the components of light microscopes and learn how to view a
sample.

Procedure:

Know the parts and components: Eyepiece- (ocular lens) what you look through to see the
sample

Arm or Neck- it provides structural support that connects head to the base.

Objective Lense- lens that provide different magnification levels to see the sample. 0 is the
smallest magnifies by 5x, 10x objective lens= 10 times magnified, 100xs is 100xs magnified.

Stage- platform that the sample is placed in order to see it through the eyepiece.

Stage clips- they secure the slide on the stage

Course focus- larger knob used to adjust the focus of the microscope, used for high
magnification

Fine focus- smaller knob used illuminate the sample for clearer viewing

Base- provides support and stability at the lower part of the microscope.

2. Place the sample on the microscope: Put the slide with the sample on the stage and
use stage clips to keep it in place.

3. choose what magnification you need- begin with lowest magnification to locate the
sample easier. Once located move the magnification up for a more detailed look.

4. adjust to optimize the view- use the course focus to get a better look at the sample.
Use the fine focus to make the small adjustments for a clearer image. Adjust the diaphragm
to regulate the light intensity for optimal clarity and brightness.

Basic idea behind microscopy

Similar to magnifying glass (needs similar components)- lens, object, and light.

How to see the sample:

Power on the light source, adjust the knob to help dim it or add more light.

Visualization: type of objectives- dry- 4, 10, and 40x. No other medium is needed to view the
object.

Oil= 100x, oil is needed for imaging. This gives better resolution on higher mags. Add a drop
of immersion oil, helps with light refraction.

When finding small tissue or bacteria start at a higher mag.

When you are uncertain start at the lowest magnification.



Results: The magnification = the power of the objective in the eyepiece.
