ILT 3 Report GMO plasmid DNA isolation 2022/2023
Goal:
The purpose of this laboratory practice was to try and isolate the plasmid which was given to us, these
GMOs were E. Coli or more precisely, DH5 with pcDNA 3.1(+) empty vector.
Our goal was to try and isolate this pcDNA 3.1(+) and by using restriction enzymes also trying to cut this
DNA and hoping that this was visible in the gel electrophoresis.
Results:
In the 3rd lane you can see the isolated pcDNA3.1 (+) ,which has a lot of large DNA bands and maybe a
few or a very low amount of small DNA bands. (The glass was not very clear).
In the control digest pcDNA 3.1 (+) (Lane 4) you can see very nice that is has been cut as there are more
smaller DNA bands and no large DNA bands.
In the isolated and digested pcDNA 3.1 (+) (Lane 5) you can clearly see both details of the control digest
and the isolated pcDNA 3.1 (+). So, this was done successfully.
