Samenvatting EM hoorcollege 1
Who needs an EM?
Earth science
Material science
Industrial manufacturing
Forensic science
Biomedics
Cell biology
Structural biology
There are some key similarities between LM and EM when you compare the microscope
subunits (illumination, lenses and detection). But there are also some key difference between
LM and EM
1. Photons are substituted with electrons
2. Glass lenses are substitutes with electromagnetic and electrostatic lenses
3. All electrons are colourless
4. In EM you have a (super) high magnification (>1.000.000x)
5. In EM you have a (extremely) small field of view (< 1 um)
Correlation magnification and small field of vieuw: the more you zoom in, the less you
see.
What is more important: high magnification or high resolution?
The resolution or resolving power: the ability to separate points of an object that are
located at a small angular distance.
Magnifiying power: the ability of the microscope to increase size
The best is to have both, but the resolution doesn’t change when you magnify so you need
both.
