Lab Report 1

Principle of Methods: The purpose or objective of this lab is to familiarize ourselves with

basic lab equipment that we will be using for the rest of the semester in lab. We will be

understanding how to accurately use and safely operate things such as a Micropipette, centrifuge,

and spectrophotometer. The experiment begins by taking a large-sized micropipette (enough to

hold 800 µl), A micropipette is a transfer device used to transfer very small amount of liquids,

hence the name micro. There is a different range set for each pipette used for different scenarios,

each with its corresponding tip to hold the liquid. The amount of liquid dispensed is based on the

number on the micropipette, this can be adjusted by turning the knob just below the plunger for

both Denville and Eppendorf. The pipette should be held firmly with all fingers wrapped around

the pipette with the thumb on the plunger. Now to aspirate or take up liquid in your pipette with

the corresponding tip on tightly, first simply press on the plunger until a small bit of resistance is

felt, this is the first stop (we do not go to the second stop because this will aspirate more than the

amount shown on the outside of the micropipette). Once that is done slowly move the pipette

down and into the desired liquid until the tip is barely submerged (about 0.5 an inch) then slowly

release the plunger and the liquid will be aspirated. For dispensing everything is the same but

there are a few minor tweaks, first lower the pipette tip close to the liquid you are dispensing into

but not in the liquid because this will cause cross-contamination. Then instead of releasing the

plunger, the plunger is pressed down first to the first stop and then to the second stop to ensure

that all of the liquid is out. After this is done eject the pipette tip into waste, which is done by

pressing the button below and to the side of the plunger. This process is done with the
