Title: PL09 Unknown Pathogen Analysis
Objective: To identify an unknown pathogen(s) based on the given assays
developed during this course.
Procedure:
Notes:
Two samples from the hospital was brought to your lab for diagnosis. After culturing
the samples on LB agar plates another co-worker began the analysis. However, due
to an emergency your co-worker has left the remaining work to you. Before leaving
your co-worker informs you the 2 liquid culture samples were accidentally combined
before the Gram stain was started. However, the 2 cultures remain separated on LB
plates for future tests.
Goal: To correctly identify the 2 pathogens.
Results:
Gram stain = Shows two samples, one stained pink and one stained purple.
One sample is spherical shaped (purple – gram positive).
The other sample is a rod shaped (pink – gram negative).
MacConkey Plates
Culture A = No growth observed.
Culture B = Lactose fermentation present, there are single colonies isolated
in zone 3. E.coli culture growth is gram negative; only gram negative will
grow on plate.
BAP Plates
Culture A = Growth present clearing around the streak; Beta hemolysis
Culture B = Growth present but no clearing bacteria, no beta hemolysis. Has
a white film and red part of the agar has changed color due to the lyses of
the red blood cells.
EMB Plates
Culture A = No growth observed
Culture B = Growth observed with green tiny that indicates it’s able to
ferment lactose; E.coli sample
Catalase Test
Culture A = Catalase positive, bubbles present, staph
Culture B = Catalase positive, bubbles present, staph
