Class notes CHEM 1127
The type of interaction between the stationary phase, mobile phase, and substances in the mixture is the main element influencing the separation of molecules from one another. Small molecules like amino acids, carbohydrates, and fatty acids can be distinguished and identified very effectively using partition-based chromatography techniques. However, the separation of macromolecules like proteins and nucleic acids can be accomplished more effectively using affinity chromatography. Alcohol, esters, lipids, and amino groups can all be separated using gas-liquid chromatography. It can also be used to track enzyme interactions. The molecular weights of proteins are particularly determined using molecular-sieve chromatography. In research on protein production, proteins are separated using paper chromatography.
